IMMUNOHISTOCHEMICAL PROCEDURE
REQUIRED SUPPLEMENTAL MATERIAL:
1. 3% H2O2: 50 mL 3% H2O2 and 200 mL 80% methanol (final concentration = 0.3% H2O2).
2. PBS
3. Ethyl Alcohol: histology grade
4. Methanol: histology grade
5. Acetone: histology grade
6. Xylene: histology grade
7. Mounting Medium
8. Hematoxylin: Gill #1
9. Goat Anti-Mouse Biotin
10. Streptavidin-HRP: Dilute in PBS and let stand for 15 minutes before use.
11. DAB: Prepare about 15 minutes before use. (WARNING: DAB is a possible carcinogen; avoid contact with skin.)
12. Normal Goat Serum (10% in PBS)
SAMPLE PREPARATION PROCEDURE:
FROZEN SECTIONS
(cut at 8 microns): Follow procedure below except frozen or fresh cut sections are air dried for 30 minutes at room temperature, fixed in acetone for 2-3 minutes, air dried for 30 minutes at room temperature.
SMEARS:
Follow the same as frozen sections procedure except fix with methanol for 30 minutes at 4C.
CYTOSPINS:
Cell lines and other cell populations are prepared using 40,000 cells in 500 mL of 10% Normal Goat Serum per well. Spin for 6 minutes at 1200 RPM. Air dry for 30 minutes at room temperature, fix in acetone for 2-3 minutes for tumor cells, or in 100% methanol for 20 minutes for peripheral blood preps, and air dry for 30 minutes at room temperature.
PARAFFIN-EMBEDDED TISSUE SECTIONS:
Place slides in a 62°C oven for 15 minutes then in Xylene for 15 minutes. De-paraffinize sections through 2 minute sequential baths of: 50% Xylene/50% ethyl alcohol; 100% ethyl alcohol; 100% ethyl alcohol; 95% ethyl alcohol; 90% ethyl alcohol; 80% ethyl alcohol; 70% ethyl alcohol; 70% ethyl alcohol. Place slides in 0.3% H2O2 for 30 minutes.
STAINING PROCEDURE:
1. Place slides in running cool tap water for 5 minutes.
2. Shake off excess water and place slides on incubation racks; circle sections with PAP pen or diamond pen to identify.
3. Flood sections with 10% Normal Goat Serum (NGS) for 15 minutes.
4. Shake off excess NGS, flood sections with primary antibody dilution, and incubate for 35-40 minutes.
5. Flood sections with PBS for 2 minutes.
6. Wipe off excess PBS and flood sections with diluted Goat Anti-Mouse Biotin for 35-40 minutes.
7. Flood sections with PBS for 2 minutes.
8. Wipe off excess PBS and flood sections with streptavidin for 10 minutes or with ABC for 35-40 minutes.
9. Flood sections with PBS for 2 minutes.
10. Wipe off excess PBS and flood sections with diluted DAB for 6-8 minutes.
11. Place slides in cool running tap water for 5 minutes, in Gill #1 hematoxylin for 90 seconds, and back in cool running tap water for 5 minutes.
12. Paraffin-embedded slides are de-hydrated through 2 minute sequential baths of: 70% ethyl alcohol, 80% ethyl alcohol, 90% ethyl alcohol, 95% ethyl alcohol, 95% ethyl alcohol, 100% ethyl alcohol, 100% ethyl alcohol, 50% Xylene/50% ethyl alcohol, Xylene, Xylene.
13. PSA reactivity may also be found in BPH (benign prostatic hypertrophy), though the pattern of staining is different than that found in carcinoma tissues. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Its protein product is a protease present in seminal plasma. It is thought to function normally in the liquefaction of seminal coagulum, presumably by hydrolysis of the high molecular mass seminal vesicle protein. Serum level of this protein, called PSA in the clinical setting, is useful in the diagnosis and monitoring of prostatic carcinoma.
