应用说明 The antibody is intended for the detection of human and murine bcl-w by Western blot, immunoprecipitation and FACS analysis.
It should be noted that the levels of antibody quoted are intended to serve as a guide only. The optimal concentration should be determined empirically by the end user.
PROTOCOLS
Western blot
In Western blot analysis, it is recommended that the antibody be used at a concentration of
1-10mg/mL, which has been shown to detect endogenous bcl-w in 30mg of total protein from mouse brain extract, or 1x106 Hela cells.
Immunoprecipitation.
For immunoprecipitation of endogenous levels of bcl-w from cell lines or tissue, it is recommended that the antibody be used at a concentration of 1-5mg/mL. It is necessary to use a minimum of 1x107 cells or 1mg of total protein from a tissue extract.
FACS.
For FACS analysis of bcl-w overexpressing cell lines, it is recommended that the antibody be used at a concentration of 1mg/mL. Expression of this gene in cells has been shown to contribute to reduced cell apoptosis under cytotoxic conditions. TISSUE SPECIFICITY: Expressed (at protein level) in a wide range of tissues with highest levels in brain, spinal cord, testis, pancreas, heart, spleen and mammary glands. Moderate levels found in thymus, ovary and small intestine. Not detected in salivary gland, muscle or liver. Also expressed in cell lines of myeloid, fibroblast and epithelial origin. Not detected in most lymphoid cell lines.
DOMAIN: SwissProt: Q92843 The BH4 motif seems to be involved in the anti-apoptotic function. & The BH1 and BH2 motifs form a hydrophobic groove which acts as a docking site for the BH3 domain of some pro-apoptotic proteins. The C-terminal residues of BCL2L2 fold into the BH3- binding cleft and modulate pro-survival activity by regulating ligand access. When BH3 domain-containing proteins bind, they displace the C-terminus, allowing its insertion into the membrane and neutralizing the pro-survival activity of BCL2L2.
