Cross-species grafting between human and animal cells or tissues requires the identification and localization of the donor material in the host environment. 应用说明 Immunohistochemistry/cytochemistry: 4% PFA fixed cryosections, or 5-10 minuted fixed cell cultures. Acetone fixation is also effective. Permeabilization with 0.1-0.4% triton X-100 is suggested in the blocking buffer. Dilution will vary slightly with lot; use at 1:10-1:500.
Formalin fixed, paraffin embedded tissues: 1:20 for 1hr at RT. Citrate buffer epitope retrieval method recommended.
Identifies an 82 and 70 kD protein by immunoprecipitation.

Chromosomes are negatively stained in metaphase cells. Also stains nuclei from primate (Sanchez-Pernaute, 2005). The antibody does not react with nuclei from mouse or rat. 同种型 IgG1 品种反应性 Human Only Species Reactivity Note The human nuclei monoclonal reacts specifically with human cells. It shows no reactivity against mouse, rat, chicken in immunohistorchemistry.
