    背景信息 CD86 (B70/B7-2) is together with CD80 (B7/B7-1) natural ligands for CD28 and CD152 (CTLA-4). It is a 70 kDa transmembrane glycoprotein belonging to the immunoglobulin gene superfamily. The CD86 cDNA encodes a 323 amino acid polypeptide composed of an N-terminal signal peptide, a 223 amino acid extracellular segment, a 23 amino acid transmembrane domain, and a 61 amino acid cytoplasmic domain (Azuma, 1993). The extracellular domain contains eight sites for N-linked glycosylation and the intracellular region has three potential sites for protein kinase C-dependent phosphorylation. The extracellular portion contains a V-set and a C2-set immunoglobulin-like domain (Azuma, 1993; Engel, 1994).
    CD86 is expressed on resting monocytes and dendritic cells and on activated T, NK, and B cells (Azuma, 1993; Engel, 1994; Caux, 1994; Engel, 1995). CD86 is rapidly induced on the surface of activated B cells. This suggests that it is the major CD28 ligand and may play an important role for costimulation of T cells in the primary immune response (Azuma, 1993; Thompson, 1995).

This protein is expressed by antigen-presenting cells, and it is the ligand for two proteins at the cell surface of T cells, CD28 antigen and cytotoxic T-lymphocyte-associated protein 4. Binding of this protein with CD28 antigen is a costimulatory signal for activation of the T-cell. Binding of this protein with cytotoxic T-lymphocyte-associated protein 4 negatively regulates T-cell activation and diminishes the immune response. Alternative splicing results in two transcript variants encoding different isoforms. Additional transcript variants have been described, but their full-length sequences have not been determined.
