背景信息	The alpha subunit of the enzyme named Mitochondrial trifunctional protein (TFP, HADHA), or alternatively TP-alpha or 78 kDa gastrin-binding protein and encoded by the human gene named HADHA/HADH is part of a mitochondrial localized trifunctional enzyme involved in fatty acid metabolism. HADHA is the alpha subunit of the holoenzyme TFP and it possesses two enzymatic activities: a long-chain enoyl-CoA hydratase and a long chain 3-hydroxyacyl-CoA dehydrogenase. HADHA is located in the inner mitochondrial membrane and HADHA catalyzes three out of the four steps in fatty acid beta-oxidation with a special affinity for long chain fatty acids. Defects in HADHA are the cause of TFP deficiency, LCHAD deficiency and maternal acute fatty liver of pregnancy (AFLP) that can be quite severe in mothers with affected fetuses. In addition, HADHA activity appears to be important in preventing breast cancer but the underlying mechanisms are just being studied. What is clear is that breast cancer tumors express low levels of HADHA and down regulation is associated with a greater risk of developing breast cancer. In other cancers it is the overexpression of HADHA that appears to be beneficial in providing resistance to certain chemotherapy drugs such as Cisplatin in lung cancer patients, and thus HADHA is being studied as a potential marker for predicting the usefulness of such drugs on patients having high or low levels of HADHA expression within their tumors. EMD-Millipore’s Anti-HADHA rabbit polyclonal antibody has been tested in against mouse kidney extracts and in paraffin embedded immunohistochemistry on human colon cancer tissue sections.
