背景信息	The renin receptor is a single pass membrane protein with a variety of older names, including ATPase H(+)-transporting lysosomal accessory protein 2, ATPase H(+)-transporting lysosomal-interacting protein 2, ER-localized type I transmembrane adaptor, Embryonic liver differentiation factor 10, N14F, Renin/prorenin receptor, Vacuolar ATP synthase membrane sector-associated protein M8-9, ATP6M8-9, and V-ATPase M8.9 subunit, each reflecting the importance of renin and prorenin to angiogenesis, blood flow and organism homeostasis. ATP6AP2 is expressed by a variety of tissues including the brain, heart, placenta, liver, kidney and pancreas, and barely detectable in lung and skeletal muscle, but present in vascular smooth muscle where renin’s action has been the most studied. Renin breaks down angiotensinogen in the blood into angiotensin I which is further broken down by ACE into angiotensin II, the primary vasoactive-vasoconstrictive peptide. When renin binds ATP6AP2, its receptor located on various tissues, the binding actually increases renin’s ability to convert surrounding angiotensinogen to angiotensin I by a factor of 4 and also induces a rapid phosphorylation of the receptor on serine and tyrosine residues associated with an activation of MAP kinases ERK1 and ERK2, which depending upon the cell type, leads to cell motility or cytoskeletal remodeling as well as other MAPK mediated events. Thus ATP6AP2 has a dual role in providing a mechanism for direct renin affects on localized cells as well as providing an increased supply of angiotensin II for the angiotensin II dependent effects needed for fluid volume homeostasis. EMD-Millipore’s Anti-ATP6AP2 rabbit polyclonal antibody has been tested in western blot on human retinal pigment epithelium tissue extracts and mouse eye tissue extracts.
