背景信息 Pttg1, also known as Securin, or Esp1-associated protein, Pituitary tumor-transforming gene 1 protein (hPTTG), or Tumor-transforming protein 1, and encoded by the gene PTTG1/EAP1/PTTG/TUTR1, is a key regulatory protein that operates in several important cellular pathways surrounding cell division as well as chromosome stability and DNA repair. Pttg1 acts by blocking the action of key proteins. During the mitosis, it blocks Separase/ESPL1 function, preventing the proteolysis of the cohesin complex and the subsequent segregation of the chromosomes. Additionally, Pttg1 negatively regulates TP53 transcriptional activity and interacts with Ku and connects DNA damage response pathways to sister chromatid separation during mitosis. Pttg1 is localized to the cytoplasm and nucleus. Pttg1 is expressed in all tissues except adult testis and up regulated in many cancer and tumor tissues.
