背景信息	The protein named Pleckstrin or alternatively Platelet 47 kDa protein (p47) and encoded by the human gene named PLEK/P47 is a major protein target for PKC found in platelets, monocytes, macrophages, lymphocytes, and granulocytes. PLEK makes up to 1% of the total protein in these cell types. PLEK is a modular platelet protein consisting of N- and C-terminal pleckstrin homology (PH) domains, a central dishevelled egl10 and pleckstrin (DEP) domain and a phosphorylation region. Following agonist-induced platelet stimulation, dimeric pleckstrin translocates to the plasma membrane, is phosphorylated by PKC and then monomerizes where it affects platelet aggregation/secretion. PLEK is also found in other cells such as phagocytes and its phosphorylation by PKC is significantly upregulated in phagocytes of individuals with diabetes mellitus compared to normal controls. PLEK appears to be involved in RAGE signaling in phagocytes and the production of proinflammatory cytokines. Overexpression of PLEK can affect ployphosphoinositide second messenger based signaling and in platelets lacking PLEK there is a severe detect in exocytosis, actin assembly, and aggregation after PKC stimulation, thus indicating multiple roles for PLEK in cellular functions. EMD-Millipore’s Anti-PLEK rabbit polyclonal antibody has been tested in western blot on U937 cell extracts and in paraffin embedded immunohistochemistry on human lung cancer tissue sections.
