背景信息	Paired box protein Pax-8 (PAX8) is a developmentally specific transcription factor governing thyroid specific gene expression and is critical for maintaining the functional and differentiated state of thyroid cell types. PAX8 is expressed developmentally in the excretory system (kidney development) and during the development of the thyroid, as well as in the adult thyroid gland. PAX8 is strongly expressed in the follicular cells of the thyroid. Defects in PAX8 expression are the primary cause for hypothyroidism. PAX8 interacts with another thyroid specific transcription factor, NK2/TTF-1 which serves as an enhancer of thyroid specific gene expression. Expression of PAX8 is confined to the nucleus as expected. PAX8 expression is also highly expressed in various cancers including kidney and ovarian carcinomas, and thyroid, renal, Müllerian, Wilms’ and thymic tumors and is often used as an identifying marker for those cancers. In stem cells, PAX8 expression particularly when paired with is enhancer partner NK2, governs the expression of at least four thyroid specific genes NIS, TSHR, Tg and TPO in multipotent endodermal stem cells. EMD-Millipore’s Anti-PAX8 rabbit polyclonal antibody has been tested in western blot on SKPV-3 cell lysates and in paraffin embedded immunohistochemistry on human ovarian tumor tissue sections.
