背景信息	The protein named DNA topoisomerase 1 and encoded by the human gene named TOP1 is the enzyme that plays the essential role in unwinding DNA necessary for DNA replication and transcription. TOP1 does so by inducing single-strand breaks via transesterification at the target site in duplex DNA. TOP1 requires ATP for breakage of the ssDNA and is specifically inhibited by camptothecin, a plant alkaloid with anti-tumor activity. Expressed in all cells but highly expressed in rapid dividing or highly expressing cells such as endothelial cells and liver cells. In disease, auto-antibodies against TOP1 are indicative of scleroderma autoimmune disease. Besides DNA TOP1 also interacts with a variety of proteins necessary to perform its primary function including BTBD1, P53, Nucleolin, BTBD2, ASF/SF2, and UBE2I. TOP1 is often up-regulated in cancers.
