背景信息 SIRT6 is one of seven mammalian homologs of the yeast Sir2 histone deacetylase. SIRT6 is a nuclear, chromatin-associated protein that is very important in normal DNA repair and resistance to DNA damage. SIRT6-deficient mice develop severe abnormalities that seem to be closely related to models of normal aging-associated degeneration and die at 4 weeks. SIRT6 has also been associated with other aging-related pathways and may be responsible for normal longevity and homeostasis. Members of the sirtuin family are characterized by a sirtuin core domain and grouped into four classes. The functions of human sirtuins have not yet been determined; however, yeast sirtuin proteins are known to regulate epigenetic gene silencing and suppress recombination of rDNA. Studies suggest that the human sirtuins may function as intracellular regulatory proteins with mono-ADP-ribosyltransferase activity. The protein encoded by this gene is included in class IV
of the sirtuin family. Alternative splicing results in multiple
