背景信息	The protein currently named GDH/6PGL endoplasmic bifunctional protein is a cytoplasmic endoplasmic reticulum enzyme expressed in all cells, but particularly high in liver cells. It has two functional domains, a glucose 1-dehydrogenase domain and a 6-phosphogluconolactonase domain. Also called Hexose-G-phosphate dehydrogenase and 6-phosphogluconolactonase (6PGL) and encoded by the human gene named H6PD or GDH the enzyme oxidizes glucose -6-phosphate, glucose, and other hexose-6- phosphate sugars and plays a critical role in generating reducing power in the cell. H6PD catalyzes the first two reactions of the pentose phosphate pathway in the endoplasmic reticulum (ER), thereby generating reduced NAD (NADPH) within the luminal compartment. Hexose-6 phosphate dehydrogenase (H6PD) is localized to microsomes and its activity also regulates corticosteroid metabolism in the liver and adipose tissue; variability in corticosteroid metabolism within the normal range have been associated with risk factors for cardiovascular disease, thus H6PD is a target of research for syndromes such as Metabolic Syndrome, and recently mutations linked to multiple sclerosis (MS) have been associated with the H6PD gene, so H6PD may be a new risk for MS patients now too. EMD-Millipore’s Anti-H6PD rabbit polyclonal antibody has been tested in western blot on HepG2 cells and in paraffin embedded immunohistochemistry on human liver cancer tissue sections.
