应用说明 Western blot: 1:2,000-1:4,000 for brain and 1:1,000-1:2,000 for muscle. The antibody recognizes a specific band at ~100 kDa. Heat the sample to 37°C in standard Laemmli for 30 minutes (don't boil). Boiling causes some sodium pump heteromers to clench together giving a band at ~150 kDa. Suggested dilution buffer is TBST with 1% BSA containing a preservative. Suggested gel percentage is 7.5%.

Na+/K+ -ATPase is an integral membrane protein responsible for establishing and maintaining the electrochemical gradients of Na and K ions across the plasma membrane. These gradients are essential for osmoregulation, for sodium-coupled transport of a variety of organic and inorganic molecules, and for electrical excitability of nerve and muscle. This enzyme is composed of two subunits, a large catalytic subunit (alpha) and a smaller glycoprotein subunit (beta). The catalytic subunit of Na+/K+ -ATPase is encoded by multiple genes.
