描述 Anti-Nociceptin Antibody, pain 背景信息 Nociceptin (Orphanin FQ) is a 17 amino acid peptide that resembles opioid peptides. Its receptor (ORL1 [Opioid Receptor-Like-1]) is expressed widely in the nervous system, indicating that this peptide participates in a broad range of physiological and behavioral functions At the cellullar level, nociceptin has potent inhibitory actions in widespread areas of the central nervous system. The nociceptin receptor uses guanine-nucleotide-binding proteins to mediate the inhibition of adenyly cyclase, the activation of potassium channels and inhibition of calcium channels.Intracerebroventricular administration of nociceptin impairs locomotion and affects nociceptin sensitivity in mice. There are data suggesting that nociceptin is a potent modulator of synaptic plasticity within the hippocampus.Spinal administered nociceptin has anti-allodynic and anti-hyperagesic effects in animal models of tonic or chronic pain from nerve injury coupled with an apparent lack of motivational effects have important implications for novel therapies involving nociceptin. 产品信息 格式 Serum 演示 Rabbit serum. Rabbit anti-Nociceptin (AB5114) Immunohistochemistry Protocol:
The Nociceptin antibody has been found to work well with the following protocol.
1. Excise tissue (myenteric plexus layer)
2. Fix in Zamboni's solution. O/N
3. Clear tissue (3 X 10' DMSO, 3 X 10' PBS) {required for good penetration}
4. Dissect tissue
5. Primary antibody diluted in antibody diluent :(4.25g NaCl; 0.2675g Na2HPO4; 0.0975g NaH2PO4.2H2O in 250 mL H2O, pH 7.1) O/N {this high salt buffer is recommended for best results}
6. Wash 3 X10' PBS
7. Secondary antibody Donkey anti rabbit CY3
8. Wash 3 X10' 50% buffered glycerol
9. Wash 3 X10' 100% buffered glycerol
10. Mount tissue on slide.
Zamboni's Fixative:
2.0% paraformaldehyde (from 16% stock solution)
15% picric acid (saturated aqueous solution)
0.1M sodium phosphate buffer stock
Adjust to pH 7.3 using 10N NaOH
Fixative is used at room temperature.
Solution may be stored at room temperature for up to six months.

Buffered Glycerol:
0.5m Sodium Carbonate Buffer, pH 8.6 + 2 Parts Glycerol
SOLUTION A-0.5M Na Carbonate Na2CO3=53g/L ~ph11.5
SOLUTION B-0.5M Na Bicarbonate NaHCO3=42g/L ~ph8.3
SOLUTION C-glycerol
Make 100 mL Soln. A 5.3g Na2CO3 in 100 mL dH2O
200 mL Soln. B 8.4g NaHCO3 in 200 mL dH2O
To make 150 mLs of Buffered Glycerol:Take 50 mL of Soln.B. Add slowly dropwise Soln. A until pH is 8.6. Use 50 mL of this mixture and add 100 mL of glycerol and mix well
