背景信息 The ADAMTS (A Disintegrin and Metalloprotease with Thrombospondin motifs) family of secreted proteases comprises at least 20 members in mammals, and several members in invertebrates. ADAMTS members resemble the membrane-bound ADAM family of proteases in the presence of a prodomain, a catalytic domain and a disintegrin-like domain. However, ADAMTS members lack a transmembrane domain, and instead contain varying numbers of thrombospondin type I modules (Tang, 2001). ADAMTS-4 was first purified by its ability to cleave aggrecan (Tortorella et al., 1999), a major cartilage proteoglycan that is degraded in patients with osteoarthritis and rheumatoid arthritis. ADAMTS-4 cleaves aggrecan between Glu373 and Ala374, a physiological cleavage site in arthritic cartilage. In addition, expression of ADAMTS-4 is elevated in chondrocytes treated with inflammatory cytokines interleukin-1, tumor necrosis factor and oncostatin M (Koshy et al., 2002; Tortorella et al., 2001). Activation of the aggrecanase activity of ADAMTS-4 requires cleavage of the prodomain by a furin-like protease and the C-terminal domain by an as-yet-unidentified matrix metalloproteinase (Gao et al., 2002).
