    特异性 The ADAM (A Disintegrin and Metalloprotease) family of cell surface proteases mediate a number of cellular processes, including differentiation, cell fusion, cell-cell interactions, and shedding of plasma membrane-bound growth factor precursors1. Each ADAM contains a prodomain, a catalytic metalloprotease domain, a disintegrin domain, a cysteine-rich domain, a transmembrane domain, and a cytoplasmic domain1. ADAM12 was first reported to be expressed by skeletal muscle cells, where it was given the trivial name meltrin-alpha. ADAM12 is transiently expressed during maturation of myoblasts to myocytes, and functions in promoting cell fusion2. In addition to the full-length form, a secreted form of ADAM12 is generated by alternative splicing3. HB-EGF has been identified as a substrate for the sheddase activity of ADAM124. Breast carcinoma cells express ADAM12, and the extracellular domain of ADAM12 is an adhesive substrate for tumor cells5. A mouse lacking ADAM12 has been generated and displays abnormalities in myogenesis and adipogenesis6. The rabbit anti-ADAM12 cytoplasmic domain recognizes a 120 kD species in lysates of MCF-7 breast carcinoma cells, which likely represents the full-length form. In addition, several lower molecular weight species, which are likely cleaved forms, are observed. EDTA is required in the lysis buffer to inhibit autolysis during lysate preparation.

Members of this family are membrane-anchored proteins structurally related to snake venom disintegrins, and have been implicated in a variety of biological processes involving cell-cell and cell-matrix interactions, including fertilization, muscle development, and neurogenesis. This gene has two alternatively spliced transcripts: a shorter secreted form and a longer membrane-bound form. The shorter form is found to stimulate myogenesis.

    DOMAIN: SwissProt: O43184 The cysteine-rich domain supports cell adhesion through syndecans and triggers signaling events that lead to beta-1 integrin-dependent cell spreading. In carcinomas cells the binding of this domain to syndecans does not allow the integrin-mediated cell spreading. & The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.
    PTM: The precursor is cleaved by a furin endopeptidase (By similarity).
