    背景信息 Glutamate receptors are the predominant excitatory neurotransmitter receptors in the mammalian brain and are activated in a variety of normal neurophysiologic processes. This gene product belongs to the kainate family of glutamate receptors, which are composed of four subunits and function as ligand-activated ion channels. The subunit encoded by this gene is subject to RNA editing (CAG->CGG; QR->R) within the second transmembrane domain, which is thought to alter the properties of ion flow. Alternative splicing, resulting in transcript variants encoding different isoforms, has been noted for this gene.

    应用说明 Immunohistochemistry:
    Dilutions of 1:10 – 1:500 were tested on rat brain lysate.
    Can be used for immunocytochemistry using paraformaldehyde or paraformaldehyde/glutaraldehyde fixed tissue with light and electron microscopy. Cryostat and vibratome sections can be used with or without Triton X-100 treatment. Suggested concentrations start at 1-3 μg/mL.

    Western blot analysis can be done at final concentrations of 1-3 μg/mL.

    Optimal working dilutions must be determined by end user.

Binding of the excitatory neurotransmitter L- glutamate induces a conformation change, leading to the opening of the cation channel, and thereby converts the chemical signal to an electrical impulse. The receptor then desensitizes rapidly and enters a transient inactive state, characterized by the presence of bound agonist.

    PTM: Palmitoylated. Depalmitoylated upon glutamate stimulation. Cys-611 palmitoylation leads to Golgi retention and decreased cell surface expression. In contrast, Cys-837 palmitoylation does not affect cell surface expression but regulates stimulation-dependent endocytosis (By similarity).
