    概述 Human recombinant histone H3.3 K27M (lysine 27 to methionine mutant) expressed in E Coli. and purified by Fast Protein Liquid Chromatography (FPLC).
    背景信息 The core histones, H2A, H2B, H3, and H4, form a central octameric structure that binds approximately 147 bp of DNA to form a nucleosome. The nucleosome is the first order of DNA compaction in the nucleus, and is the basic unit of chromatin. The structure of chromatin can be altered by post-translational modifications (PTM) occurring on the C- and N-terminal tails of H2A, H2B, H3, and H4. Histone modifications can either provide or hinder access of key transcriptional complexes to DNA sequences and can regulate the expression of genes. These effects can be deciphered by applications such as chromatin immunoprecipiation (ChIP), which requires high quality chromatin, and essential tools such as histone proteins and appropriate enzymes, to assemble chromatin in vitro. Mutations of lysine 27 (K27) to methionine (M), have been associated with the development of pediatric glioblastoma (see Lewis, P. W. et al. Inhibition of PRC2 activity by a gain-of-function H3 mutation found in pediatric glioblastoma. Science 28 Mar 2013).
