背景信息 Rho-Associated Kinase (Rho-Kinase or ROK) has been classified into two isoforms: ROKα/ROCK-II and ROKβ/ROCK-I. These large cellular proteins (150-160 kDa), have been reported to be involved in the formation of stress fibers, focal adhesions and cardiac hypertonic responses. Current research suggests that ROK of smooth muscle may be involved in the contractile process via phosphorylation of MYPT1 and Myosin. The serine-threonine kinase ROCK-1 is a down-stream effector of Rho, and is involved in signal transduction pathways that regulate cell morphology, motility, and tumor progression through the reorganization of the actin cytoskeleton. 产品信息 格式 Affinity Purified 控制 - A431 cell lysate

Entrez基因汇总 This gene encodes a protein serine/threonine kinase that is activated when bound to the GTP-bound form of Rho. The small GTPase Rho regulates formation of focal adhesions and stress fibers of fibroblasts, as well as adhesion and aggregation of platelets and lymphocytes by shuttling between the inactive GDP-bound form and the active GTP-bound form. Rho is also essential in cytokinesis and plays a role in transcriptional activation by serum response factor. SUBUNIT STRUCTURE: Binds RHOA (activated by GTP). Interacts with ADD1, GEM, RHOB, RHOC, MYLC2B and VIM By similarity. Binds RHOE, PPP1R12A, LIMK1 and LIMK2. Interacts with TSG101. Interacts with DAPK3.

PTM: Autophosphorylated on serine and threonine residues. Phosphorylated upon DNA damage, probably by ATM or ATR.
Cleaved by caspase-3 during apoptosis. This leads to constitutive activation of the kinase and membrane blebbing.
