背景信息 Interleukin 22 Receptor alpha 1 (IL-22RA-1) is a member of the type II cytokine receptor family and has recently been discovered to be produced by Th17 cells. IL-22RA-1 and IL-10RB form a complex that mediates IL-22 signaling via JAK/STAT pathways. Current studies have revealed that IL-22 targets cells of the digestive, skin, and respiratory organ systems and plays an important role in mucosal immunity. The IL-22 receptor (IL-22R) is expressed exclusively in these tissues, thereby allowing the cytokine to mediate epithelial innate immunity in response to a variety of pathogens. Although recent studies have shown the importance of IL-22 in host defense against Gram-negative bacterial organisms (in gut and lung), there is evidence that IL-22 also plays a role in autoimmune disease, such as psoriasis. UniProt汇总 FUNCTION: Component of the receptor for IL20, IL22 and IL24. Component of IL22 receptor formed by IL22RA1 and IL10RB enabling IL22 signaling via JAK/STAT pathways. IL22 also induces activation of MAPK1/MAPK3 and Akt kinases pathways. Component of one of the receptor for IL20 and IL24 formed by IL22RA1 and IL20RB also signaling through STATs activation. Mediates IL24 antiangiogenic activity as well as IL24 inhibitory effect on endothelial cell tube formation and differentiation.
SUBUNIT STRUCTURE: Heterodimer with IL10RB and with IL20RB. IL22 binding to heterodimer is greater than binding to IL22RA1 alone.
SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein.
TISSUE SPECIFICITY: Expressed in colon, liver, lung, pancreas and kidney. No expression in immune cells such as monocytes, T-cells, and NK-cells. Expressed in keratinocytes of normal skin as well as in psoriatic skin lesion. Detected in normal blood brain barrier endothelial cells as well as in multiple sclerosis lesions; Strongly expressed on central nervous system vessels within inflitrated multiple sclerosis lesions. Overexpressed in synovial fluid cells from rheumatoid arthritis and spondyloarthropathy patients.
INDUCTION: By interferon-gamma in keratinocytes.
MISCELLANEOUS: Failure of medical and surgical therapy in Chronic rhinosinusitis with nasal polyps is associated with decreased expression of IL22RA1.
