背景信息 SHPS-1 (also known as P84, BIT, and SIRP) is a neuronal membrane glycoprotein that promotes the attachment and neurite outgrowth of cultured murine cerebellar cells. Because of its heterophilic adhesive properties and its localization at synaptogenesis sites, SHPS-1 is suggested play a role in the regulation of synapse formation or maintenance. As a member of a phosphatase binding protein family, SHPS-1 shares identical homology with murine SHPS-1 cDNA. It is involved in receptor tyrosine kinase signaling that is found at synapses in the mammalian central nervous system and in non-neural tissues. It is reported that integrin-associated protein (IAP/CD47) is a predominant binding partner for P84. Upon phosphorylation of its cytoplasmic region, SHPS-1 binds to the protein-tyrosine phosphatase SHPS-2. Formation of the SHPS-1-SHP-2 complex is also implicated in the regulation of cellular migration. SHPS-1 and its ligand CD47 constitute an intercellular recognition system that contributes to the inhibition of cell migration via cell-cell contact. The ectodomain of SHPS-1 is shown to be excised from cells in a reaction that is likely mediated by a metalloproteinase. It is demonstrated that forced expression of a SHPS-1 mutant resistant to ectodomain shedding impaires cell migration, cell spreading, actin cytoskeleton reorganization, and paxillin- and FAK- tyrosine phosphorylation. 产品信息 格式 Purified 控制 - Mouse brain tissue lysate

UniProt汇总 FUNCTION: Immunoglobulin-like cell surface receptor for CD47. Acts as docking protein and induces translocation of PTPN6, PTPN11 and other binding partners from the cytosol to the plasma membrane. Supports adhesion of cerebellar neurons, neurite outgrowth and glial cell attachment. May play a key role in intracellular signaling during synaptogenesis and in synaptic function. Involved in the negative regulation of receptor tyrosine kinase-coupled cellular responses induced by cell adhesion, growth factors or insulin. Mediates negative regulation of phagocytosis, mast cell activation and dendritic cell activation. CD47 binding prevents maturation of immature dendritic cells and inhibits cytokine production by mature dendritic cells.
SUBUNIT STRUCTURE: Binds PTPN11 when tyrosine-phosphorylated, except in macrophages, where it primarily binds PTPN6. Binds GRB2 vitro. Binds FGR. Binds JAK2 irrespective of its phosphorylation status and forms a stable complex. Binds SCAP1 and/or SCAP2. The resulting complex recruits FYB. Binds PTK2B.
SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein.
TISSUE SPECIFICITY: Highly expressed in cerebral cortex, brain, spinal cord, cerebellum and spleen, and at much lower levels in kidney, thymus, heart, lung and liver. Within the cerebellum, highly expressed throughout the molecular layer, and in synaptic glomeruli in the granule cell layer. Detected in neurons of the hippocampus and dentate gyrus, and in olfactory bulb. Not detected in Purkinje cells. Highly expressed in the plexiform layers, optic fiber layer and the outer segments of the photoreceptor layer in the retina. Highly expressed in macrophages.
