背景信息	CoA synthase, or also known as Bifunctional coenzyme A synthase, NBP, or POV-2 and encoded by the human gene named COASY is the enzyme that catalyzes the two final stages of de novo coenzyme A (CoA) biosynthesis. COASY enzyme has two functional domains of activity, a phosphopantetheine adenylytransferase activity, or alternatively named Dephospho-CoA pyrophosphorylase, Pentetheine-phosophate adenylyltransferase (PPAT) activity, and a Dephospho-CoA kinase activity (DPCK) or alternatively Dephosphocoenzyme A kinase (DPCOAK) activity. Because of the dual activities COASY is thought to be a key point for the regulation of CoA biosynthesis. The enzyme is a monomer and expressed by all tissues in moderate amounts, and while expressed in the cytoplasm it is always associated with mitochondria. Lowest levels are seen in the lymphocytes and highest in the kidney. COASY does have isoforms, with isoform 2 being found expressed in the brain. COASY interacts in vitro with Src homology-2 (SH2) domains of a number of signaling proteins, including Src homology-2 domains containing protein tyrosine phosphatase (Shp2PTP) and p85alphaPI3K. Recent research also indicates that COASY interacts with several other kinds of intracellular proteins including scaffold proteins like EDC4 (enhancer of mRNA-decapping protein 4) which can influence one or more of COASY biosynthetic activities, thus regulating the biosynthesis of coenzyme A. EMD-Millipore’s Anti-COASY rabbit monoclonal antibody has been tested in western blot on HepG2, HeLa, and A431 cell lysates and by flow cytometry on HeLa cells in culture.
