描述 FlowCellect™ Human FOXP3 Treg Characterization Kit 概述 We have developed a multi-parameter flow cytometry assay for the characterization of human regulatory T-cells. Millipore’s FlowCellect™ Human FOXP3 Treg Characterization Kit includes three directly conjugated antibodies: CD3 PE/Cy5, CD4 FITC, and FOXP3 Alexa Fluor® 647, along with optimized fixation, permeabilization, wash, and assay buffers to provide researchers the ability to phenotypically distinguish human regulatory T-cells. Our combination of directly conjugated antibodies and optimized buffers allow for detection of CD3 and CD4 on the surface as well as FOXP3 within the nucleus simultaneously.
Antibodies against CD3 and CD4 are provided in the kit to characterize cells by their surface marker expression. CD3 is a marker for the T-cell receptor, while CD4 is a glycoprotein that is found on T helper cells, monocytes, dendritic cells, macrophages, and regulatory T-cells . When looking at CD4 positive cells which are also positive for CD3 you are effectively eliminating the Monocytes, Macrophages, and dendritic cells present in peripheral blood or PBMC’s from your analysis. These cells which are CD3+, CD4+, and also FOXP3 + identify the regulatory T-cell population.Alternate Names - Flow Cellect
背景信息 Regulatory T-cells (Tregs) are a subpopulation of helper T-cells which are CD3+, CD4+, CD25+, and express the transcription factor FOXP3. Characterization of these cells can be accomplished by performing multi-parameter flow cytometry to identify key combinations of the aforementioned surface and intracellular markers.
Regulatory T-cells play a vital role in regulating the responses of the immune system. They suppress the activation of the immune system following infections, which helps maintain a balance or homeostasis. It has also been shown that some cancer patients have increased numbers of regulatory T-cells which may allow some malignant cells to escape the activation of the immune system due to the suppressive nature of the Tregs. Researchers are interested in the mechanism by which cancers are evading responses of the immune system as these could aid in the design of alternate treatment methods.
A second and equally important function of regulatory T-cells is promoting tolerance to self-antigens. Regulatory T-cells act to ensure that self-antigens presented to the immune system do not illicit an immune response. This is yet another way that regulatory T-cells help to maintain homeostasis.
Many researchers are interested in the area of self-tolerance as a way to identify what can be tolerated and what signals a response to antigens. In particular, it would be of great benefit to understand the immune response that is common in organ, tissue, and cellular transplantation from donors to recipients.
