背景信息	The small ADP Ribosylation Factor (ARF) GTP-binding proteins are major regulators of vesicle biogenesis in intracellular traffic. ARF proteins cycle between inactive GDP-bound and active GTP-bound forms that bind selectively to effectors. ARFs regularly associate with two types of protein, those involved in catalyzing GTP/GDP exchange (GEFs), and those that serve other functions. Cytohesin-2 (CYTH2) or alternatively named ARF exchange factor, ARF nucleotide-binding site opener (ARNO), PH, SEC7 and coiled-coil domain containing protein 2 and encoded by the human gene named CYTH2/ARNO/PSCD2 and PSCD2L acts as a guanine-nucleotide exchange factor (GEF) and promotes the GTP/GDP exchange between ARF1, ARF3 and ARF6. Found localized in the cytoplasm and membrane, CYTH2 is recruited to the cell membrane only when associated with ADP-ribosylation factor-like proteins (ARL’s), ARL4A, ARL4C, and ARL4D. CYTH2 and its related proteins regulate protein sorting and membrane trafficking which play critical roles in everything from cellular homeostasis to outgrowth and movement. For instance in neurons, ARF6, CYTH2 and actinin-1 operate together to regulate neurite extension. In cancer, CYTH2 expression appears to be an important indicator of survival. CYTH2 is over expressed in human lung cancer and appears to activate ErbB cytoplasmic receptor mediated signaling. CYTH2 promotes HepG2 proliferation through the IGF pathway, and CYTH2 also promotes VEGF-dependent initiation of angiogenesis vessel permeability endothelial proliferation in various tumors. EMD-Millipore’s Anti-CYTH2 rabbit polyclonal antibody has been tested in western blot on mouse heart tissue lysates and in paraffin embedded immunohistochemistry on human ovarian tumor tissue sections and in fluorescent immunocytochemistry on HeLa cells in culture.
