背景信息 The protein coded by the gene named JMJD2B is Lysine-specific demethylase 4B protein. It is also known as JmjC domain-containing histone demethylation protein 3B or Jumonji domain-containing protein 2B with gene names JMJD2B, JHDM3B, or KIAA0876. The Jumonji (JMJ) protein family plays important roles in the development of many tissues. The JMJD2A-D proteins are a family of histone demethylases that contain a conserved domain termed a Jumonji C (JmjC) domain. This entire family is associated with cancer-associated genes principally because JMJD2C is amplified in esophageal squamose cell carcinoma (ESCC) JMJD2B specifically demethylates 'Lys-9' of histone H3, thereby playing a role in histone controlled gene expression, chromatin modification, and epigenetics. The JMJD2B protein is specific for Lys-9 methylations on histone H3 and does not demethylate histone H3 'Lys-4', H3 'Lys-27', H3 'Lys-36' nor H4 'Lys-20'. The protein can only demethylate trimethylated H3 'Lys-9', and it does so with less vigor than other known demethylases. Interestingly, JMJD2B’s expression is upregulated in hypoxia suggesting a governing role in hypoxic gene expression as well as chromatin modification and epigenetics.
