背景信息	Soluble cytosolic carbonic anhydrases (CAs) are well known to participate in pH regulation of the cytoplasm of mammalian cells. Membrane-bound CA isoforms, such as isoforms IV, IX, XII, XIV, and XV, also catalyze the reversible conversion of carbon dioxide to protons and bicarbonate, but at the extracellular face of the cell membrane. Carbonic anhydrase 9 or alternatively Carbonate dehydratase IX, Carbonic anhydrase IX,(CA-IX,CAIX), Membrane antigen MN, P54/58N, Renal cell carcinoma-associated antigen G250, RCC-associated antigen G250, or pMW1 and encoded by the human gene named CA9/G250/MN is one of the CA membrane bound enzymes that metabolizes CO2 into carbonic acid and thus helps mediate intracellular pH. CA9 expression is typically limited to the surface of microvilli but is also found in the nucleus and in particular the nucleolus. Expression in normal tissues is restricted primarily to the gastric mucosa as one would expect, however CA9 is also found in other cells such as neurons and is highly expressed in many cancer cells, and CA9 is the only tumor associated carbonic anhydrase isoenzyme known. The expression of CA9, at least in cancers, seems to be driven by hypoxia via HIF1 and it is possible that HIF1 or other cellular stress indicators drive the expression of CA9 in neurons and normal cells as well. CA9 expression is being studied as a biomarker of survival in hypoxic tissues and in tissues recovering from trauma such stroke or severe injury. EMD-Millipore’s Ant-CA9 rabbit polyclonal antibody has been tested in western blots on HEK293 cell lysates and in paraffin embedded immunohistochemistry on human liver cancer tissue sections.
