背景信息 C4d is a degradation product of activated Complement 4 (C4), a central component in the classical complement cascade pathway. This pathway is typically activated by specific antibody binding to select target molecules, making C4d an indirect footprint or marker of antibody response. After cleavage occurs, C4d is left with several exposed ester groups that allow for C4d to bind in a transient, covalent manner to endothelial cell surfaces along with components of ECM vascular basement membranes near C4 activation sites. Due to the strength of these bonds and Cd4’s ability to act as an indirect marker of antibody response, this degradation product has become a significant marker, both general and robust, for humoral rejection of transplants including allografts from cardiac, kidney, and liver tissues. It has also been shown to bind platelets associating Cd4 as a marker for both stroke severity and ischemic stroke (Mehta, et al., 2008). 产品信息 格式 Purified 控制 - Aged mouse serum

UniProt汇总 FUNCTION: C4 plays a central role in the activation of the classical pathway of the complement system. It is processed by activated C1 which removes from the alpha chain the C4a anaphylatoxin. The remaining alpha chain fragment C4b is the major activation product and is an essential subunit of the C3 convertase (C4b2a) and the C5 convertase (C3bC4b2a) enzymes of the classical complement pathway.
SUBUNIT STRUCTURE: Circulates in blood as a disulfide-linked trimer of an alpha, beta and gamma chain.
SUBCELLULAR LOCATION: Secreted.
PTM: Prior to secretion, the single-chain precursor is enzymatically cleaved to yield the non-identical chains (alpha, beta and gamma). During activation, the alpha chain is cleaved by C1 into C4a and C4b, and C4b stays linked to the beta and gamma chains. Further degradation of C4b by C1 into the inactive fragments C4c and C4d blocks the generation of C3 convertase.
MISCELLANEOUS: C4 is a major histocompatibility complex class-III protein.
