背景信息	The protein Thymidine kinase, cytosolic, and encoded by the human gene TK1 is the enzyme that combines thymidine and ATP to make deoxythymidine 5’ phosphate, the beginning compound needed to make thymidine triphosphate needed for DNA synthesis. TK1 activity is highest during cell proliferation and low in resting cells. It is expressed in all cells and also highly expressed in cancer cells. TK1 is present in the cytoplasm only and it forms a tetramer when fully activated by ATP. TK1 activity is regulated by thymidine triphosphate levels which serve as a negative feedback regulator. Normally TK1 is synthesized during cell division and does not pass into the blood for body fluids except if cellular damage occurs. In metastasis of certain cancers, TK1 levels in the serum is often used as a biomarker for cancer growth rate in such cancers. Also, because TK1 is expressed only during S-phase it is also often used as a marker of cell division and as a general cancer cell marker in conjunction with other markers. TK1 in the serum is also used as a data point for diagnosis of non-malignant diseases too such as vitamin B12 deficiency and pernicious anemia, various viral infections including Herpes, and deep wound or trauma injury. EMD-Millipore’s Anti-TK1 rabbit polyclonal antibody has been tested in western blots on HEK293 cell extracts and in fluorescent immunocytochemistry on HeLa cells in culture.
