背景信息	The protein named Ubiquitin carboxyl-terminal hydrolase 1 or alternatively, Deubiquitinating enzyme 1, (hUBP), Ubiquitin thioesterase 1, or Ubiquitin-specific-processing protease 1 and encoded by the human gene named USP1 is one of many hydrolases that removes ubiquitin from specific proteins. USP1 is special because it specifically interacts and deubiquitinates key enzymes that are critical to the proper function of DNA repair when ubiquitinated. FANCD2 and PCNA are proteins involved in DNA repair synthesis that normally are monoubiquitinated to form a scaffold for DNA repair enzymes. USP1 deubiquitinates those thus preventing proper DNA repair. USP1 requires a cofactor, WDR48 for highest activity. USP1 is cytoplasmically localized. More recent research also shows that USP1 can play a role in regulating kinases such as those involved in PI3K/AKT pathway, and down regulation of USP1 increases PIK3/AKT activity, and such activity has been linked to aggressive tumor formation, and such cancers often have very low USP1 activity, thus augmentation of USP1 in cancer may be beneficial in some forms of cancer. EMD-Millipore’s Anti-USP1 rabbit polyclonal antibody has been tested in western blot on HEK293 ce ll lysates and in paraffin embedded immunohistochemistry on human cervical cancer tissue sections.
