背景信息	The protein called THAP1 or THAP domain-containing protein 1, or Thanatos-associated domain containing apoptosis-associated protein 1 and encoded by the human gene named THAP1 is a DNA-binding transcription regulator that regulates endothelial cell proliferation and G1/S cell-cycle progression. THAP1 specifically binds the 5'-[AT]NTNN[GT]GGCA[AGT]-3' core DNA sequence and acts by modulating the expression of pRB-E2F cell-cycle target genes, including RRM1. THAP1 is a component of the THAP1/THAP3-HCFC1-OGT complex that is required for the regulation of the transcriptional activity of RRM1. THAP1 may also have pro-apoptotic activity because when overexpressed it potentiates both serum-withdrawal and TNF-induced apoptosis. Additionally, the THAP domain co-localizes with the apoptosis response protein PAWR/PAR-4 in promyelocytic leukemia (PML) nuclear bodies and appears to be the functional link between PAWR and PML nuclear bodies in that disorder. THAP1 is localized in the nucleus and expressed in a variety of tissues most highly in skeletal and cardiac muscle but THAP1 expression is also found in kidney, brain, and liver. THAP1’s gene is also associated with the DYT6 gene of Dystonia 6, a hereditary movement disorder involving sustained, involuntary muscle contractions. EMD-Millipore’s Anti-THAP1 rabbit polyclonal antibody has been tested in western blot on HEK293 cell extracts and in paraffin embedded immunohistochemistry on human colon cancer tissue sections.
