背景信息 Polycomb group ring finger 2, also known as PCGF2, is a transcriptional repressor. Polycomb group (PcG) of genes were first identified in Drosophila whereby these genes were required to sustain the stable repression of Hox cluster genes during development. Studies have shown that PCGF2 forms multimeric protein complexes associated with chromatin. In addition, PCGF2 may play a role in cell proliferation and/or neural cell development. PcG gene products form complexes via protein-protein interaction and maintain the transcription repression of genes involved in embryogenesis, cell cycles, and tumorigenesis. This protein was shown to act as a negative regulator of transcription and has tumor suppressor activity. The expression of this gene was detected in various tumor cells, but is limited in neural organs in normal tissues. Component of the Polycomb group (PcG) multiprotein PRC1 complex, a complex required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility.
SUBUNIT STRUCTURE: Exists as both a monomer and homodimer By similarity. Component of chromatin-associated class II polycomb repressive complex 1 (PRC1/hPRC-H) at least composed of PCGF2/RNF110, BMI1/PCGF4, CBX2/M33, CBX4/PC2, CBX8/PC3, PHC1, PHC2, PHC3, SCMH1, RING1 and RNF2/RING2.
SUBCELLULAR LOCATION: Nucleus.
TISSUE SPECIFICITY: Detected in all tissues examined with high expression found in placenta lung and kidney and low expression, in liver, pancreas and skeletal muscle.
PTM: Phosphorylated. Homodimer formation is regulated by phosphorylation state with unphosphorylated protein forming homodimers By similarity.
