    背景信息 Programmed cell death protein 4 (PDCD4) is widely studied for its role as a tumor suppressor. It inhibits the translation of RNA molecules indirectly, by binding to the eukaryotic initiation factor (elF) proteins that are involved in unwinding the secondary structure of RNA molecules, such as the p53 mRNA, before translation. PDCD4 tightly binds, via its C-terminal MA-3 domains, to elF4A and elF4G and consequently blocks protein synthesis and potential cell growth and proliferation, which suppress further development of transformed cells. PDCD4 is regulated by post-translational modification and proteosome degradation. It is phosphorylated on serine 67 by the S6K1 kinase and degraded by the SCF ubiquitin ligase. A number of studies have indicated that PDCD4 is downregulated in various human carcinomas.
