概述	The Fragile X Syndrome is identified by an ampiflication of the FMR1 (CGG)n repeat region. The Fragile X CHEMI™ probe is a digoxigenin-labeled DNA probe (pFxa1NHE) which can be used in conjunction with the Sure Blot® CHEMI™ Kit (Cat. No. S4250) to detect (CGG) repeats in the FMR-1 gene. The same results are obtained as when using the standard P32-labeled probes, but without radioactivity. The amplification of the FMR-1 gene is detected on Southern blots by an increase in the band sizes. Also provided is a ready-to-use mixture of unlabeled and digoxigenin-labeled Lambda DNA/Hind III Molecular Weight Markers for convenient visualization on both ethidium bromide-stained agarose gels and Southern blots.

For best results, target human genomic DNA should be double digested, to completion, with the restriction enzymes EcoR I and either Sac II, Nru I, or Eag I. S1120-1 Fragile X (pFxa1NHE) DNA Probe Locus: FMR1 Volume: 250 μl Concentration : 10ng/μl; 25 μl per 10 ml of hybridization solution. Sufficient probe to make 100 ml of hybridization solution at a probe concentration of 25 ng/ml. (equal to processing 1500 cm2) Restriction Map: see insert. S1120-2 Lambda/Hind III Molecular Weight Markers Fragile X CHEMI™ Molecular Weight Markers are Lambda DNA cut with Hind III. Each vial contains a ready-to-use mixture of unlabeled and digoxigenin-labeled DNA fragments in gel loading solution. The unlabeled markers are included for convenient visualization on agarose gel after staining with ethidium bromide. The fragment sizes are: 23.1, 9.4, 6.6, 4.4, 2.3, 2.0, 1.0, 0.56, 0.125 kb. The 0.125 kb fragment may be difficult to detect, and will likely electrophorese off the end of the gel. Volume: 800 μl. Sufficient for 20 lanes, 10 gels with 2 lanes per gel, 40 μl per lane. Instructions for Use

Molecular Weight Markers

1. Thaw contents of S1120-2 at room temperature or 37°C and aliquot 40 μl per lane into a separate microcentrifuge tube.

2. Incubate the microcentrifuge tube containing the aliquoted markers at 65°C for 3-5 minutes. Load 40 μl per lane onto an agarose gel and proceed with electrophoresis and transfer.

Do not incubate the stock tube of markers at 65°C.

Probe Hybridization

For complete hybridization, detection, and stripping protocols please refer to the S4250 instruction manual.

Procedural Cautions

Temperature and buffer salt concentration (stringency) of hybridization and washing are important, as lower stringency can result in non-specific binding of the probe to other sequences, and higher stringency can result in lack of signal.
