背景信息	Pituitary homeobox 3, also named Homeobox protein PITX3 or Paired-like homeodomain transcription factor 3 with genes names PITX3 or PTX3 is a homebox transcription factor that is essential for the differentiation and maintenance of the meso-diencephalic dopaminergic (mdDA) neurons during development and for their long term survival and maintenance during adulthood. Several variants of this gene have been associated with human Parkinson's disease (PD), and lack of PITX3 in mice causes the preferential loss of substantia nigra pars compacta (SNc) mdDA neurons that are most affected in PD. Thus the study of PITX3 expression and the genes it helps mediate is of critical importance in understanding the pathology of Parkinson’s disease. PTX3 activates NR4A2/NURR1 mediated transcription of many genes essential for mdDA growth and survival including SLC6A3, TH, and DRD2. Interestingly PITX3 also plays important roles in the eye where it is essential for proper lens development and fiber cell differentiation and the induction of specific crystalline proteins. Defects in the gene are the cause of numerous cataract or developmental eye related abnormalities. Highly expressed in the developing eye and dopaminergic regions in the brain, PITX3 is was recently shown to be a GDNF responsive gene that is essential for activating the expression of brain-derived neurotrophic factor (BDNF) in mdDA neuron subpopulations during embryogenesis. The loss of BDNF expression is correlated with the increased apoptotic cell death of mdDA neuronal subpopulations in mice lacking PITX3. The regulation of GDNF, PITX3, and BDNF expression also persists in the adult brain, and this suggests that any impairment or disruption of the regulatory interaction between these three factors may contribute to the loss of mdDA neurons and subsequent disease pathology.
