背景信息	Presenilin is a probable catalytic subunit of the gamma-secretase complex, an endoprotease complex that catalyzes the intramembrane cleavage of integral membrane proteins such as Notch receptors and APP (beta-amyloid precursor protein). Presenilin requires the other members of the gamma-secretase complex to have a protease activity. It may play a role in intracellular signaling and gene expression or in linking chromatin to the nuclear membrane. The gamma-secretase complex stimulates cell-cell adhesion through its association with the E-cadherin/catenin complex. Under conditions of apoptosis or calcium influx, it cleaves E-cadherin promoting the disassembly of the E-cadherin/catenin complex and increasing the pool of cytoplasmic beta-catenin, thus negatively regulating Wnt signaling. Futher researcher may implicate the complex in hematopoiesis. Defects in PSEN1 have been implicated in early-onset Alzheimer disease type 3 (AD3) and late-onset Alzheimer disease (LOAD).

应用说明	Immunocytochemistry: A previous lot of this antibody was used in IC.Acetone or methanol fixed cells; or lightly fixed 2.5-3% PFA (5-10' RT). Immunohistochemistry: A previous lot was used on 3% paraformaldehyde fixed tissue. Works on paraffin embedded tissue sections if fixed with PLP, acetone or other light fixations; 10% formalin fixation is untested. Immunoprecipitation: A previous lot of this antibody was used in IP. These disease-linked mutations result in increased production of the longer form of amyloid-beta (main component of amyloid deposits found in AD brains). Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor, such that they either directly regulate gamma-secretase activity or themselves are protease enzymes. 存储条件	Stable for 6 months at -20ºC in undiluted aliquots from date of receipt. Handling Recommendations: Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
