背景信息 Bromodeoxyuridine (BrdU) is a thymidine analog and is specifically incor-porated into DNA during DNA synthesis. Anti-bromodeoxyuridine monoclonal antibody is used to identify cells that have incorporated BrdU. This immunological detection scheme has several advantages over the use of radioactive thymidine incorporation for identifying cells under-going replication. Labeling and detection can be performed the same day instead of waiting several days, as required for autoradiography of tritium-labeled cells, and the necessity of using multiple specimens for obtaining the optimal exposure time is eliminated. In addition, anti-bromodeoxyuridine staining with flow cytometric analysis allows multiple parameters to be evaluated simultaneously. 产品信息 格式 Culture Supernatant 控制 - Brdu treated cells
演示 UnPurified mouse tissue culture supernatant from a perfusion system, filtered through a 0.2μ micron membrane prior to vialing. Product contains 20%FBS and Ciprofloxacin at final concentration of 10μg/mL. Possible mycoplasma risk contained within this product. 应用说明 Immunocytochemistry: 1:5000-1:10,000; acid/alcohol fixed cells and tissues (5 parts glacial acetic acid and 95 parts of 95% ethanol; BU-1 may react with traditional fixations, however it has not been closely examined.
PROTOCOL FOR SLIDE-BASED LABELING INDEX ASSAY USING MAB3510
Procedure:
1. Assay performed on mononuclear cells isolated from whole blood or bone marrow using Ficoll™ density gradient.
2. Cytospin slides are prepared using 2.5-5.0 x 104 cells per slide (microscope slides - 1x3 inch, PTFE coated, Cell-Line Associates, Inc. #10-1179).
3. Cells are fixed in 95% Ethanol for 10 minutes at room temperature.
4. Wash slide with PBS containing 0.05% Tween 80.
5. Add 100 mL of MAB3510 diluted appropriately (if PTFE-coated slides are used, the antibody will stay in place over the cells).
6. Incubate for 30 minutes at room temperature.
7. Wash slide with PBS/Tween.
8. Add 100 mL of goat anti-mouse conjugated to Rhodamine (AP124R) appropriately diluted in RPMI containing 10% fetal calf serum.
9. Incubate for 30 minutes at room temperature.
10. Wash slides with PBS/Tween and allow to dry.
11. Place a drop of n-propyl gallate on the cell spot and coverslip.
12. Store slides in the dark at 2-8°C until read.
Flow Cytometry: 1:10,000 (95% ethanol fixed cells tested)

MAB3510 does not require pretreatment of cells with strong DNA denaturants in order for the antibody to recognize incorporated BrdU.
