背景信息	Fumarate hydratase (FH) or alternatively known as Fumarase is an enzyme that catalyzes the reversible conversion of fumarate to malate. Fumarase comes in two forms, a mitochondrial form and a cytosolic form. FH is the mitochondrial form and it is a key enzyme in the Krebs cycle. The cytosolic form of fumarase is involved mostly in amino acid metabolism. Defects in FH activity lead to severe neurologic abnormalities and anemia. FH is expressed in most tissues, and being part of the Krebs cycle, it is found in the mitochondrial matrix. FH is also thought to be a tumor suppressor, although it is the cytosolic form that appears to be the major provider since only it can migrate to the nucleus and play a role in the DNA damage response. Mutations in FH are useful markers for hereditary leiomyomatosis and renal cell cancer syndrome (RCC). Down regulation of FH is related to tumorigenesis. The loss of expression correlates well with the progression from normal parenchyma cells to RCC, thus FH may be an indicator of tumorigenesis in sporadic RCC. Finally, FH in the mitochondria and FH in the cytosol appear to be under different controlling mechanisms. FH in the mitochondria for instance is increased during periods of under nutrition, whereas the cytosolic form is not. Additionally, the cytosolic form is increased in certain reward pathways in the brain of rats upon stimuli whereas the mitochondrial form is not and its level appears to be controlled by other mechanisms. EMD-Millipore’s Anti-FH (mitochondrial form) rabbit polyclonal antibody has been tested in western blot on HeLa cell extracts and in paraffin embedded immunohistochemistry on human prostate cancer sections and in fluorescent immunocytochemistry on HepG2 cells in culture.
