应用说明 Immunofluorescence and immunoperoxidase staining of formalin-fixed and paraffin-embedded sections of anterior pituitary glands. Cryostat sections of the same tissues can also be used.
Optimal working dilutions must be determined by the end user.
IMMUNOPEROXIDASE STAINING:
1. Hydrate the sections with PBS, pH 7.4.
2. Incubate the section with antiserum diluted 1:1000 in PBS at 37°C for 30 minutes.
3. Wash for 15 minutes with PBS at room temperature and treat sequentially with Goat anti-Rabbit IgG and PAP complex at 37°C. Wash excess reagent from the section as described above.
4. Treat the section with a mixture of 3,3'-diaminobenzidine HCl (0.02%) in hydrogen peroxide (0.004%). For preservation, the sections may be dehydrated, cleared and mounted.
IMMUNOFLUORESCENCE STAINING:
1. Same as Step 1 above.
2. Incubate the section with antiserum diluted 1:20-1:40 in PBS at 37°C for 30 minutes.
3. After washing off the excess antiserum with PBS, treat the section with fluorescein labeled Goat anti-Rabbit IgG for 30 minutes at 37°C. Wash the tissue with PBS and mount under glycerine-saline.生物信息 宿主 Rabbit 特异性 Gives a positive and specific immunostaining of FSH-containing cells.
