背景信息	The protein named Interferon-inducible protein AIM2 or “Absent in melanoma 2” protein and encoded by the human gene named AIM2 is another member of the family of proteins seemingly involved in human innate response. AIM2 appears to be involved in innate immune response by recognizing cytosolic double-stranded DNA and inducing caspase-1-activating inflammasome formation in macrophages. AIM2 binding and oligomerization triggers activation of caspase-1, and the processing and release of IL-1beta and IL-18 from macrophages. AIM2 can recognize dsDNA of viral or bacterial origin in a non-sequence specific manner and trigger PYCARD dependent caspase-1 cell death, thus AIM2 is a double-stranded DNA receptor. AIM2 also appears to be a tumor suppressor operating via apoptosis activation pathways and the repression of NF-kappa-B transcriptional activity and subsequence control of cell proliferation. AIM2 is found primarily in the nucleus but also is found in the cytoplasm and expressed by a wide variety of cell types and leukocytes. AIM2 expression is induced by interferons and defects in AIM2 function may be the cause of unstable microsatellite DNA seen in certain cancers such as colon cancer, and because of its role in the activation of host defense, it is possible that AIM2 also contributes to autoinflammatory or autoimmune diseases.
