背景信息	The protein named Zinc finger protein GLI2, or alternatively the Tax Helper protein, and encoded by the human gene GLI2/THP is transcriptional activator that is involved in mediating Sonic hedgehog signaling pathway activities. GLI2 also augments Tax-dependent gene enhancer functions and plays a role during embryogenesis. GLI2 is expressed in the cytoplasm but relocates to the nucleus and binds DNA via the classic zinc finger protein motif. In development , GLI2 affects ventroposterior mesodermal development through its regulation of Wnt, Brachyury and Xhox3 gene families. Knockout studies suggest that GLI1 can compensate for GLI2 but replacement is not perfect and is context dependent. GLI2 null mice embryos develop neural tube defects, and double knockout GLI1 and GLI2 mice display serious central nervous system and lung defects. Overexpression of GLI2 causes tumors and GLI2 is expressed in the interfollicular epidermis and the outer root sheath of human normal skin. Recent studies have shown that inappropriate activation of GLI2 can lead to basal cell carcinomas. Thus, GLI2 is being studied as both a neurodevelopmental agent and as a possible cancer target for skin cancers in humans. EMD-Millipore’s Anti-GLI2 rabbit polyclonal antibody has been tested in western blots on HEK293 cell extracts and in paraffin embedded immunohistochemistry on human kidney tissue sections.
