背景信息 The protein Cyclin D2 (CCND2) functions as the regulatory component of the CDK4, CDK5 and CDK6 kinases, which are critical kinases that govern the transition of the cell through G1/S checkpoint. CCND2’s normal function is to activate the CDK enzymes which in turn allow the cell cycle to proceed. Without activation the CDKs do not phosphorylate their targets and cells are held at the checkpoint. Mutant CCND2 overexpression is observed in several cancers including myelomas, gastric, squamous, and prostate cancers, while in others, CCND2 expression is reduced or non-existent and reintroduction of its expression halts division. CCND2-CDK complexes also interact with the tumor suppressor protein Rb. Phosphorylation of Rb allows and activates E2F target genes and is responsible for progression through the G1 phase. Over activation of CCND2 results in more CCND2-CDK complexes and less non-phosphorylated Rb, thus the cells are pushed to divide. CCND2 is localized to the nucleus primarily but can be found in the cytoplasm/peri-nuclear area as well. CCND2 levels are partially controlled via ubiquitination and subsequent degradation, and in some forms of cancer it is the degradation of CCND2 that is defective again leading to an oversupply and continual push through the various checkpoints. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
